Studies on the Immunohistochemistry of Anisakis larvae


Abstract
The cuticle showed the negative reaction at any antibody titers. This study was performed to evaluate the comparative antigenicity of body structures of Anisakis larvae. Excretory-secretory antigens(ES-Ag), Whole worm extract antigen(WWE-Ag), and
somatic antigen(SM-Ag) of Anisakis larvae were prepared. And rabbit anti-excretory-secretory antigen antiserum(anti-ES) and rabbit anti-Anisakis antiserum(anti-An) were obtained. Indirect enzyme linked immunosorbent assay was applied to reveal
the
responsibility between two antisera and 3 antigens. After that, indirect immunoperoxidase staining were applied to show the antigenicity of sectioned Anisakis larvae.
@ES The results were summerized as follows:
@EN 1. IgG titers of two antisera were significantly increased during the immunization schedule.
2. The ES-Ag showed the highest antigenicity among three tested antigens.
3. The order of reaction intensity of three antigens to antisera was ES-Ag, SM-Ag, and WWEAg.
4. There were no differences of stained sites of sectioned Anisakis larvae except reaction intensities at indirect immunoperoxidase staining with anti-ES and anti-An.
5. The subcuticle showed the comparatively high antigenicity in the body compartments of sectioned Anisakis larvae. The intestine and esophagus were moderately stained.
6. The reaction intensities of ventral cord, dorsal cord, lateral cord, and muscle layer were weak.
7. The cuticle showed the negative reaction at any antibody titers.
